I attended the Plastic and Reconstructive Surgery Department of Kansai Medical University (Osaka, Japan) from July 22nd to August 1st, 2013. During this time I had the chance to witness the surgical procedures performed, use their laboratories and prepare the platelet gel with the use of different kits. We have set up a standardized method of preparing PRP for use in the joint research protocol. Following critical discussion of the research protocol, we decided to make the following changes:
- The study phase on the mouse model will take place at the Kansai Medical University, while the subsequent phase on humans will take place at theUniversità Campus Bio-Medico di Roma, subject to approval by the Ethics Committee.
- The Department of Plastic, Reconstructive and Aesthetic Surgery ofUniversità Campus Bio-Medico di Roma will provide Japanese colleagues with 300 mg "custom-made" prostheses for implantation in animals.
- We reduced the age of the rats at operation to 8 weeks
- Rats will be divided into 7 groups of 3 animals each. 2 prostheses will be implanted in each animal.
- We decided to focus on PRP and removed the fat graft treatment: rats in the experimental groups will be treated with high concentration PRP or low concentration PRP.
- RT-PCR for alpha-SMA and Collagen type 1 will be performed in the explanted periprosthetic capsule samples.
